摘要
α-淀粉酶抑制剂作为一种减肥和降血糖的辅助药物,越来越受到更多的关注。同时它可作为抗虫的基因,在医学和农业上应用前景广阔。该文中对小麦麸皮中α-淀粉酶抑制剂的分离、纯化工艺以及性质进行了研究。结果表明,小麦麸皮粗提物经纤维素离子交换柱和凝胶柱层析分离纯化,得到精制α-淀粉酶抑制剂,经PAGE-SDS凝胶电泳得到3条带,分子质量分别为33 ku,30 ku和26 ku。该提取物在80℃以下,pH3-10,抑制活力相对稳定。Na+,K+,Mg2+在0-200 mmol/L具有提高α-淀粉酶抑制剂活力作用,高浓度的Ca2+具有一定抑制作用。
As an ancillary drug for weight loss and hypoglycemia, α-Al gets more and more attention. It also can be used as zoophobous gene. The application of α-Al in the fields of medical science and agriculture is multifold. The technology of isolation, purification and characteristics of α-amylase inhibitor from wheat bran has been researched. The purified α-Al was obtained by ion exchange chromatography and gel filtration column chromatography. There were three bands by SDS-PAGE, the molecular was 33ku,30ku and 26ku. The analysis indicated that the activity of α-Al was stable at pH 3 and temperature under 80℃. The activity of α-Al raised when the concentration of Na^+ , K^+ , Mg2 + was under 200 mmol/L, and the high concentration of Ca2+ can inhibit the activity of α-Al.
