摘要
采用Plackett-Burman(PB)设计法筛选出影响灵芝菌G.lucidum UIM-281发酵产漆酶的主效应因素为装液量、摇床转速和培养时间。最陡爬坡实验接近以上三因素最优水平后,应用中心组合设计以及响应面分析,建立了以漆酶酶活为响应值的二次回归方程模型,得到最优发酵条件为:装液量52 mL,摇床转速158 r/min,培养时间172 h,温度28℃,接种量10%,初始pH5.5,种龄96 h。此条件下理论预测值8342.6 U/L,验证实验测得酶活为8387.7 U/L,与预测值接近,说明预测模型可靠性较高,可应用于漆酶发酵条件的优化。
The main factors effecting laccase yield of G. lucidum UIM-281 was firstly screened by Plackett-Bur- man(PB) design. After steepest ascent experiment approaching the optimal level of the three factors, the center combination design and response surface analysis were applied to establish the second-order equation model for the yield of laccase. The optimal fermentation conditions were obtained as following: 52 mL of broth content, 158 r/min of rota- ting speed, 172 h of culture time, 10% of inoculation volume, and 96 h of seed age at initial pH 5.5 and at 28℃.The verification test value of the laccase activity was 8 342.6 U/L, which was nearly in agreed with the theoretical prediction value 8387.7 U/L. The results suggested the predicted model was reliable and available for the optimization of laccase fermentation conditions.
