大鼠心血管系统中Ⅱ型磷脂酶A_2 mRNA表达及cDNA克隆

摘要 目的:探讨Ⅱ型磷脂酶A2(PLA2)mRNA在正常大鼠心血管系统中的分布情况,研究其基因和氨基酸顺序结构的特征,了解与心血管疾病有密切关系的PLA2。方法:从大鼠血、心肌、主动脉弓、下腔静脉近心段、腹主动脉和下腔静脉腹腔段的组织匀浆中提取总RNA,合成第一链cDNA,利用PCR扩增大鼠Ⅱ型PLA2mRNA,并对大鼠腹主动脉的PLA2cDNA进行克隆和序列分析。结果:在所检测的几个心血管系统组织中均能测到Ⅱ型PLA2mRNA,大鼠腹主动脉的DNA和氨基酸结构与其他组织来源的PLA2有高度同源性。结论:Ⅱ型PLA2广泛存在心血管系统中,Ⅱ型PLA2mRNA在血液和主动脉弓中的表达较高,提示这可能是一种有益的选择性表达,发挥着潜在的宿主防御细菌感染功能。 Objective:To investigate the distribution of type Ⅱ phospholipase A2 (PLA2)mRNA in rat cardiovascular system and study the eDNA and amino acid sequences of the PLA2 structure , in order to increase our knowledge in their possible role in the circulation system. Methods:Extracted total RNA from organs homogenate of cardiovascular system. Then the first-strand eDNA was synthesized and PLA2 DNA was amplified by PCR. The DNA sequence was cloned and analyzed. Results: Type Ⅱ PLA2 mRNA was detectable in blood, myocardium, aortic arch, cardiac segment of inferior vena cava, abdomial aorta and coeliae segment of inferior vena cava. The eDNA and amino acid sequence was highly homologous with other PLA2 eDNA from different tissues of the rat. Conclusion:Type Ⅱ PLA2 mRNA was found in all organs of cardiovascular system and platelet PLA2 mRNA content was rich in blood and aortic arch, which suguested that PLA2 might be beneficial selective expressin and play a potential role in the process of host defense bacteria infection and inflammation.
