摘要
【目的】了解葛根素对腹膜透析液干预下的体外培养的人腹膜间皮细胞增殖活性及转化生长因子β1(TGF-β1)分泌的影响。【方法】体外培养的人腹膜间皮细胞,分对照组、PDS组、A组(葛根素终浓度为50μg/mL)、B组(葛根素终浓度为100μg/mL)、C组(葛根素终浓度为200μg/mL)五组观察。检测各组上清液中TGF-β1的含量以及间皮细胞的增殖活性并比较。【结果】腹膜间皮细胞在PDS干预下,TGF-β1分泌显著增加,添加葛根素再干预的A、B、C组TGF-β1分泌与PDS组比较有显著下降。与PDS组比较,A、B、c纽间皮细胞增殖活性显著升高。【结论】葛根素可以抑制腹膜间皮细胞TGF-β1分泌,拮抗腹膜透析液对腹膜间皮细胞增殖活性的抑制作用。
[Objective] To investigate the effect of puerarin on the cell proliferation activity and the secretion of TGF-β1 of human peritoneal mesothelial cell(HPMC) cultured in vitro with peritoneal dialysate(PDS). [Methods] HPMCs were cultured in vitro and divided into five groups (RPMI1640 culture medium as control group, PDS containing 2.5% glucose as PDS group, PDS containing 2.5% glucose and 100μg/mL, 200μg/ mL or 400μg/mL puerarin as group A, group B and group C, respectively). After 48h incubating, mitochondrial dehydrogenase activity(MTT assay) of HPMC and the levels of TGF-β1 in supernatant were detected and compared. [Results] Significantly decreased mitochondrial dehydrogenase activity was observed in PDS group as compared with those in control group and group A, B and C( P 〈0.01). Significantly lower level of TGF- β1 in supernatant was found in control group, as well as in group A, B and C, when compared with that in PDS group( P〈 0. 01). [Conclusion]Puerarin can restore the reduced cell proliferation activity caused by PDS. Also, it can reduce the secretion of TGF-β1 induced by commercial PDS in cultured human peritoneal mesothelial cell.
