硅胶G柱层析法制备卵黄卵磷脂工艺的研究

摘要 以鸡蛋卵黄为材料,采用二元有机溶剂为提取剂,丙酮为沉淀剂,应用正交试验确定卵黄卵磷脂的最佳提取条件;选择硅胶G为吸附剂,以不同有机溶剂配比组成的洗脱体系为洗脱剂,经单因素试验柱层析法纯化卵磷脂.结果表明:提取温度25℃,提取时间60min,醇醚体积比4∶1提取效果最佳;无水乙醇、甲醇混合溶剂为最佳纯化洗脱剂组合,并获得了纯度达99.3%的卵黄卵磷脂. The yolk was used to extract lecithin. Silica- gel was used as absorbent and the orthogonal test was used to confirm the best condition. 99.3% purity of the purified lecithin was obtained by column chromatography purification. The results showed the optimal conditions were temperature 25℃, time 60 min, the ratio of alcohol ether 4 : 1 respectively. The mixture solution of alcohol and methanol were the best solvent.
