摘要 [目的]通过检测猪瘟疫苗免疫后的IFN-γ水平探讨仔猪细胞免疫情况。[方法]采用超前免疫和普通免疫2种方法对仔猪进行猪瘟免疫,用流式细胞仪对猪瘟疫苗免疫后仔猪外周血单核细胞(PBMC)中分泌IFN-γ细胞数量进行检测。[结果]无论采取哪一种方法进行免疫,仔猪PBMC中分泌IFN-γ细胞的比例均显著增多,且一直维持在较高的水平。当在60日龄仔猪进行第2次免疫后分泌时,IFN-γ细胞的比例增加更为明显,而对照组仔猪分泌IFN-γ细胞的比例在试验过程中则一直维持较低的水平,这说明猪瘟疫苗能够诱导仔猪体内IFN-γ水平升高,间接表明猪瘟疫苗注射后激发了机体的细胞免疫应答。[结论]与普通免疫方法相比,采用超前免疫方法能够使仔猪及早地产生针对猪瘟的细胞免疫反应。 [ Objective] This study aimed to discuss the immunization of the cell in piglets immunized with classical swine fever vaccine through detection on IFN-γ Level in immunized piglets. [ Method] Piglets were immunized with classical swine fever (CSF) vaccine by common immunization and ultra Pre-immunity and the cells secreting IFN-γ in peripheral blood mononuclear cells (PBMC) were detected by flow cytometry after piglet immunization. [ Result] The proportion of cells secreting IFN-γ in PBMC was increased significantly and retained a high level no matter which immunity method was adopted and that was increased more significantly after the secondary immunization for piglets at 60 days,while that in CK group retained a lower level during the whole process of experiment, indicating that the immunization of CSF vaccine could induce the rising of level of IFN-γ in piglets and indirectly indicating that the cellular immunity was provoked after the injection of CSF vaccine. [ Conclusion ] Compared with the method of common immunization, the ultra pre-immunity method could made the piglets early pro- duce the cell immunoreaction aiming to CSF.
