超声法提取竹节参总皂苷工艺条件的优化 被引量:6

摘要 [目的]为竹节参皂苷进一步开发利用提供依据。[方法]通过比色法测定竹节参总皂苷含量,考察料液比、乙醇浓度、提取温度、提取时间、超声功率等因素对竹节参总皂苷得率的影响,并采用正交试验优化超声法提取竹节参总皂苷的工艺条件。[结果]各因素对竹节参总皂苷提取效果的影响顺序为:超声功率>料液比>提取时间>提取温度;得到最佳提取工艺为:浓度70%乙醇,料液比1∶20,提取温度50℃,超声功率为32kHz,提取3次,每次30min。在该提取工艺下总皂苷得率为13.15%。[结论]优化出了超声法提取竹节参总皂苷的最佳工艺条件,该工艺具有提取温度低、时间短、得率高的优点。 [ Objective ] The basis of further development and utilization of saponin from Japanese ginseng( Panax japonicum) was provided. [ Method] The content of total saponin from Japanese ginseng was measured with the method of colorimetric deter mination. The effect of the ratio of material to solution, the concentration of ethanol, extracting temperature, time, ultrasonic power and other factors on the extraction of total saponins of Japanese ginseng was researched. And the extraction condition of total saponin with ultrasonic method was optimized with orthogonal test. [ Results] The order of various factors affecting the extraction of total saponin from Japanese ginseng was as follows: the ultrasonic Power 〉 the ratio of material to solution 〉 extraction time 〉 extraction temperature. The best extraction process was follows : the concentration of ethanol, 70% ; the ratio of material to solution, 1:20 ; the extraction temperature ,50 ℃ ; the ultrasonic power,32 kHz and the extraction time,3 times for 30 minutes for each. The rate of total saponin extracted was 13.15%. [ Conclusion] The condition of total saponin extracted from Japanese ginseng with the ultrasonic method was optimized optimum ,which advantage was low extraction temperature, short time and high yield.
