天麻素对戊四氮致癎大鼠海马代谢型谷氨酸受体1和蛋白激酶Cα表达的影响 被引量:6

Effects of Gastrodin on Expressions of mGluR1 and PKCαin Hippocampus of Rats with Seizures Induced by Pentylenetetrazole

摘要 目的:观察天麻素对戊四氮致癎大鼠海马代谢型谷氨酸受体1(mGluR1)和蛋白激酶Cα(PKCα)表达的影响。方法:成年雄性Wistar大鼠102只随机分成:对照组;戊四氮组;丙戊酸钠干预组;小剂量天麻素(100 mg·kg^(-1))干预组和大剂量天麻素(200 mg·kg^(-1))干预组。每组再分4个时间点:12h、48h、5d和7d。用免疫组化检测海马mGluR1和PKCα的变化。结果:除对照组无癎样发作外,其余各组大鼠全部出现Racine分级Ⅱ~Ⅴ级癎样的发作。rnGluR1和PKCα阳性细胞数量在戊四氮组明显增加、平均A值增加,与对照组比差异有统计学意义(P<0.05);与丙戊酸钠组比差异无统计学意义(P>0.05);与小剂量和大剂量天麻素干预组比差异有统计学意义(P<0.05)。结论:天麻素通过对戊四氮致癎大鼠海马mGluR1和PKCα信号传导通路的抑制作用,影响大鼠癎样发作。 Aim: To observe the effects of gastrodin on the expressions of metabotropic glutamate receptor 1 (mGluR1) and protein kinase Cα(PKCα) in hippocampus of rats with seizures induced by pentylenetetrazole (FTZ), and explore the role of gastrodin in the epilepsy to genesis and development process. Methods: 102 healthy adult male Wistar rats were randomly divided into 5 groups: a control group, a PTZ group and a sodium vaproate (VPA) intervening group, a gastrodin low dose (G1) intervening group and a gastrodin high dose (Gh) intervening group. Each group was further classified into 12 h, 48 h, 5 d and 7 d subgroups. The expressions of mGluR1 and PKCα in hippocampus were demonstrated with immunohistochemistry. Results: The expressions of mGluR 1 and PKCα in the hippocampal area were stronger in the PTZ group and VPA intervening group than those of the control group, and their average A values had significant difference compared with the control group. There were no significant difference in the expressions of mGluR1 and PKCα between the G1 and Gh intervening groups. And their average A values had significant difference compared with the control group. There were also significant difference compared with the FFZ group. Condusion: Gastrodin, by inhibiting the signal transduction of mGluR 1 and PKCα in hippocampus, can alleviate the seizure activities.
