肿瘤坏死因子对培养人牙周膜成纤维细胞c-fos表达的诱导作用

The study of the expression of c-fos in periodontal ligament fibroblasts induced by tumor necrosis factor

目的：深入探索肿瘤坏死因子对人牙周膜成纤维细胞的生物学功能影响的机制。方法：将５０００Ｕ／ｍｌ肿瘤坏死因子作用于培养至第三代的人牙周膜细胞３ｈ后，进行ＦＯＳ免疫组化反应。结果：几乎所有的ＰＤＬＦｓ核均染为棕黄色，为ＦＯＳ免疫组化反应阳性，对照组细胞核未见着色。结论：由于ｃ－ｆｏｓ的表达可以提示核内基因的进一步改变。

Aim: To study effects of tumor necrosis factor(TNF) on periodontal ligament fibroblasts(PDLFs). Methods: 5×10 3 U/ml TNF was added into the third generation PDLFs for 3 hours, then FOS immunocytochemical reaction and HE staining were conducted. Results: HE staining results showed that cells were of normal cell structure, clear cell outline and clear nucleus with no granules in cytoplasm. FOS immunocytochemical reaction results showed that nearly all the nuclei of PDLFs were stained dark yellow-brown, which showed FOS positive immunocytochemical reaction. Conclusions: Since that the expression of c-fos in PDLFs after TNF acting suggested that genes in nuclei changed correspondingly. This article indicated that TNF might affect the biological function of PDLFs on a deeper level.
