广东省深圳地区汉族人群MN血型表现型的特点及基因频率调查 被引量:3

摘要 目的探讨深圳地区汉族人群MN血型表现型的特点及分布情况。方法以试管法检测246倒汉族人的红细胞M,N血型抗原,流式细胞术定量检测血清学凝集反应有明显差异的样本,分析其红细胞上抗原数量的差异;并进行基因型鉴定。结果试管法定量检测中发现抗原、抗体反应凝集强度存在差异,以流式细胞术验证单个红细胞M抗原的荧光强度同样存在强弱之分,同时以SSP—PCR基因分型技术鉴定血型特异性,基因分型结果与血清学分析结果相符。深圳地区汉族人群中M,N基因频率分另日为0.4756,0.5244。结论深圳地区汉族人群MN血型分布特征基本符合中国汉族人群的分布特征,红细胞M,N血型血清学反应的个体差异是否与红细胞表面抗原数量或质量相关有待进一步探讨,同时增加样本数量对该血型系统遗传多态性进行研究。 Objective To discuss the character and distribution for the phenotype of MN blood group in han population of Shenzhen area. Methods Among 246 subjects of Han population of M and N antigens were quantitative detected by test tube technique and checked the agglomeration that obvious discrepancy in serology by flow cytometry to analyzed the antigens amount on the erythrocytes. At the same time the allele frequencies of M and N in population was detected. Results The agglutination of the serological ration examine by test tube and the mean fluorescence intensity(MFI) on a single M antigen by flow cytometry had obvious discrepancy. The MN blood group was genotyped by polymerase chain reaction-se quence specific primers (PCR-SSP). FCM and genotyping method showed consistent results with serologic testing. Gene frequencies of M and N were 0. 475 6 and 0. 524 4 respectivelyin han population of Shenzhen area. Conclusion The distribution for the phenotype of MN blood group in han population of Sbenzhen area accorded with Chinese han, It will be dis cussed the obvious discrepancy in serology for the M and N antigens of individual whether or not connect the antigens' amount with quality. At the same time ,author will continue investigation the genetic polymorphisms of MN blood group by augment samples.
