摘要
目的探讨骨髓细胞染色体制备过程中R显带技术的影响因素,分析显带液的pH值和显带时间的相关性。方法骨髓标本278例(良性血液病患者80例,恶性血液病患者198例),培养收获后,采用R显带技术,观察分析显带液的pH值与显带时间对显带效果的影响。结果显带温度87.5℃,6.5〉pH〉6.0时,随显带液pH值增高,显带时间逐渐延长,其中6.4〉pH〉6.2,显带时间65-85min时显带成功例数最多,pH〉6.5或pH〈6.0,显带常失败;良性血液病患者显带成功的比率高于恶性血液病患者。结论在影响骨髓细胞染色体R显带技术的诸多因素中,显带液pH值与显带时间对显带效果的影响最显著,6.4〉pH〉6.2,显带时间65-85min时显带效果最佳,且随显带液pH值增高,显带时间应运渐延长;良性血液病患者的显带效果优于恶性血液病患者。
Objective To investigate the influential factors for R-banding technique during chromosomal preparation in bone marrow cell and analyze the correlation between pH value of banding fluid and banding time. Methods 278 bone marrow prepares were obtained with 80 benign hemopathies and 198 malignant ones. The correlation between pH value of banding fluid and banding time were observed with R-banding technique. Results When the pH value was increased gradually and banding time rised accordingly. A relatively stable and accurate chromosome banding image could be obtained at 87.5℃ of banding temperature and 6.5〉pH〉6.0. The number of perfect banding images was maximum while banding time ranged 65-85 minutes and pH value for 6.2-6.4. The banding achievement ratio of benign hemopathies were higher than malignant ones. Conclusion The pH value of banding fluid and banding time were important compare to others in influential factors for R-handing technique. When banding time ranges 65-85 minutes and pH value for 6. 2-6.4,the banding image was excellent. The banding images of benign hemopathy was superior to the ones of malignant hemopathy.
