摘要
目的探讨枸杞与细胞凋亡以及枸杞、自由基和细胞凋亡以及运动能力和运动性蛋白尿之间的关系,旨在为运动训练提供一个有效的辅助手段,为中药在运动训练和健身中的广泛而深入的应用提供依据。方法选用50只Wistar大鼠随机等分为对照组(安静,安静力竭,训练,训练力竭)和药物组(安静,安静力竭),采用递增负荷游泳运动。测凋亡蛋白用免疫组织化学方法,显示肾脏组织中凋亡蛋白Bcl-2和Bax的分布和表达特点,用计算机图像采集与分析技术,显示各组大鼠肾脏Bcl-2和Bax表达的变化,检测β2-微球蛋白和尿白蛋白浓度。结果(1)力竭时间:实验前后训练组和药物组的力竭时间均延长,且训练组和对照组比有显著性差异(P<0.05)。(2)计算机图像观察:M1组Bcl-2免疫阳性反应明显增强,棕黄色颗粒染色密度增加,C2组Bcl-2免疫阳性反应最弱,棕黄色颗粒几乎没有;C2组Bax免疫阳性反应明显增强,棕黄色颗粒染色密度增加,M1组Bax免疫阳性反应最弱,棕黄色颗粒减少。(3)免疫组化结果:Bcl-2:与C1比较,C2、T1组Bcl-2明显降低,P<0.05,M1略有上升,但P>0.05;与C2比较,T2明显上升,P<0.05。M2稍有上升,但P>0.05;与T1比较,T2下降明显,P<0.05;与M1比较,M2下降,P<0.05。Bax:与C1比较,C2组Bax明显升高,P<0.01;与T1比较,T2升高特别显著,P<0.01;与M1比较,M2上升幅度很大,P<0.01;与C2比较,T2稍有降低,P<0.05;M2降低但不明显,P>0.05,与C1比较,T1、M1组都降低,P<0.05。(4)Alb:与C1比,T1、M1组大鼠尿液Alb含量差异不显著。与C2组比,T2组降低,且P<0.01;M2组降低,且P<0.05。(5)β2-MG:与C1比,T1、M1组大鼠尿液β2-MG含量差异不显著,无统计学意义。与C2组比,T2组大鼠尿液β2-MG浓度低,且P<0.01,M2组大鼠尿液β2-MG浓度降低,且P<0.05。结论枸杞、运动使肾脏的抗氧化、抗自由基能力明显增强,枸杞主要通过降低Bax蛋白表达的机制来解除其对抗Bcl-2蛋白抑制凋亡的作用,运动训练和枸杞可以通过调节Bcl-2/Bax比值来抑制细胞凋亡,均能降低大鼠力竭即刻状态下尿液Alb及β2-MG浓度,提高运动能力。
The author discusses the relation between the Lycium barbarmn and apoptosis, among the Lycium barbarum, free radical, exercise capability and athletic albuminuria. Furthermore, the author would like to provide an effective auxiliary method to strengthen exercise capability and an evidence to adopt Chinese medicine widely in the waning and physical exercising. Method: 50 healthy 8 - week - old female Wistar rats which weighed 220 g ± 23.19 g each were divided into six teams of CONTROL and MADICINE - TAKEN randomly. They were trained to take incremental swimming. The location and expression feature of apoptosis protein Bcl-2 and Bax were showed with immunohistochemistry. Finally, Bcl-2 and Bax in the rats' kidneys were analyzed with the computer image collection and analysis technology to demonstrate the variety of Bcl-2 and Bax protein expression in different teams. Detection of Alb and β2-MG concentration. Results: (1) exhaustive time: both before and after the experiment, the training group and drug group extended exhaustive time, and training group and contract group had significant differences( P 〈 0. 05). (2) computer image: Bcl-2 immune positive reaction of M1 groups enhanced obviously, and it's tan particles increases, while C2 group has the weakest Bcl- 2 immune positive reaction with few yellowish - brown particles; Bax immane positive reaction of C2 group enhanced obviously, the density of dyeing tan particles increased. M1 group had the weakest Bax immune positive reaction with yellowish-brown particles decreasing. (3) immunohistochemical result: For Bcl-2, comparing with C1, Bcl-2 of C2 and T1 decreased significantly, P 〈 0.05, and M1 rose slightly, but P 〉 0.05; comparing with C2, T2 increased obviously, P 〈 0.05, and M2 rose slightly, butP〉0.05; comparing with T1, T2 dropped significantly, P 〈0.05; comparing with M1, M2 dropped, P 〈 0.05. For Bax, comparing with C1, Bax of C2 increased significantly, P 〈 0.01 ; compar/ng w/th T1, T2 increased signifwantly( P 〈 0. 01 ) ; comparing with M1, M2 rose greatly, P 〈 0.01 ; comparing with C2, T2 slightly got decreased, P 〈 0.05, M2 decreased less significantly, P〉 0.05; comparing with C1, T1 and M1 decreased, P 〈 0.05. (4) Alb: difference of Alb content in rat urine of TI and MI group was not so significant. Comparing with C2, Alb concentration ratio ofT2 decreased and P 〈 0.01, and so did M2 group (P 〈 0.05). (5) β2- MG: comparing with C1, difference of β2- MG content in rat urine ofT1, M1 was not so significant with no statistical significance. Comparing with C2, theβ2 - MG density of T2 rats kept low and P 〈 0.01, and so did the M2 rats, (P 〈 0.05). Conclusion: Taking Lycium barbarum and training enhances the kidney's capacity of anti - oxidation and anti - free radical. The Lycium barbarian relieves whose antagonism Bcl-2 coming the mechanism expressing mainly by reducing the apoptosis protein Bax restrain the body' s cell apoptosis much more obviously. Furthermore, taking training and Lycium barbarum can restrain cell apoptosis and enhance the exercise capability by adjust the ratio of Bcl-2 and Bax, also can reduce rat exhaustion instantly condition of Alb and β2 - MG.
