摘要 目的:罗布麻愈伤组织的培养及发状根的诱导。方法:以罗布麻无菌种子来源的愈伤组织为材料,选出适合于诱导发状根的较硬而分生细胞较多的愈伤组织扩增的培养基,对扩增的愈伤组织进行液体培养,摸索适合诱导发状根的培养条件。结果:适合于诱导发状根的较硬而分生细胞较多的愈伤组织扩增培养基为:MS+0.5mg/L 6-BA+0.3 mg/LNAA+300mg/L LH,在24±2℃,光强度为1500-2000Lx,光照周期为12h光/12h暗下培养;在17-200C、60rpm的黑暗、换液周期为40-42d的条件下培养,适合于发状根产生的培养基为1/2MS+0.1mg/LNAA+35g/L蔗糖,低温是罗布麻发状根产生和生长的必要条件。本文的研究结果为罗布麻发状根的规模化培养体系的建立和主要次生代谢药物的代谢调控研究奠定了基础。 Objective: cultureof callusand inducement of hairy roots of Apocynum venetum. Methods: Callus derived from seeds of A. venetum were amplified on a series of mediums, optimal medium was screened for propagation of the callus which is harder and with more meristematic cells. The callus was cultured in liquid medium for exploring the conditions of hairy roots inducement. Results:MS+0.5mg/L 6-BA+0.3 mg/LNAA+300mg/L LH was the most appropriate medium for the callus propagation, culture temperature was 24 +2℃ ,illumination was 1500-2000Lx and photoperiod was 12h light/12h dark. I/2MS+0. lmg/LNAA+35g/L sucrose was the optimal liquid medium for production and growth of hairy roots,the culture conditions were at 17- 200C,rotation speed was 60rpm in dark and 40-42d was a cycle of renewing medium. Low temprature was the necessary to rooting. These results laid the foundation for the establishment of a large-scale culture system of A .venetum hairy roots and the reseaeh for metabolic control of the main secondary metabolites.
