摘要
利用生物信息学方法对来自2个不同cDNA文库的2万余条杨树EST序列进行了候选SNPs标记。研究结果表明,在获得的94个碱基替代型候选SNPs中,A/G替换类型最多,占32.98%,C/T替换型占13.83%,其它碱基置换型SNPs占53.20%。同时,对其中20个候选SNPs位点的重新测序结果表明,有13个(占65%)候选SNPs位点得到证实,其中,11个位点为碱基替代型SNPs,另外2个为杂合型位点。
More than 20 000 EST sequences of poplar were clustered and stringent filters were used to identify reliable candidate SNPs. Sequences analysis indicated that totally 94 candidate SNPs were found. The main types of substitutions among candidate SNPs were A/G, C/T and other types, which accounted for 32. 98%, 13. 83%, and 53. 20%, respectively. Resequencing with 6 samples for 20 candidate SNPs showed that 13 candidate SNPs were verified, accounting for 65 %. And among them, 11 SNIPs were substitution type and the other 2 SNPs were heterozygous type. The detailed information about these candidate SNPs would provide a basis for further marker identification and application.
