姜黄素对尤文氏肉瘤细胞系RD-ES细胞的诱导凋亡作用研究 被引量:1

Mechanism of the Curcumin-induced Apoptosis of Ewing's Sarcoma RD-ES Cells

摘要 目的:研究姜黄素对尤文氏肉瘤细胞系RD-ES细胞的诱导凋亡作用。方法:体外培养人尤文氏肉瘤RD-ES细胞,实验分为4组,即空白对照(空白溶液)及姜黄素低、中、高剂量(1、10、100mg·mL-1)组。24~72h后观察各组细胞的形态变化,并分别用MTT、TUNEL染色、RT-PCR及图像分析技术检测各组细胞生长、凋亡率及Bcl-2基因表达水平的变化。结果:与对照组比较,姜黄素低、中、高剂量组RD-ES细胞生长受阻,细胞凋亡率增加,Bcl-2表达减少。结论:姜黄素可诱导RD-ES细胞凋亡,并与Bcl-2减少有关。 OBJECTIVE: To explore the mechanism of the curcumine - induced apoptosis of Ewing' s sarcoma RD - ES cells. METHODS: The RD- ES cells were cultured in vitro and treated with blank solution (control) or curcumine solution (1, 10, and 100 mg ·mL^ -1) for 24-72 hours. The cell morphology at 24-72 h was observed, and the cell growth, apoptotic rate, and Bcl -2 gene expression were detected by MTT assay, TUNEL staining, RT- PCR and image analysis technology, respectively. RESULTS: Compared with control group, RD- ES cells treated by curcumine solution (1,10, and 100 μmol · L^-1) showed growth inhibition and increased apoptotic rate and decreased Bcl - 2 expression. CONCLUSION : Curcumine can induce apoptosis of RD- ES cells, and the mechanism is associated to Bcl - 2 reduction.
