摘要 目的:建立以高效液相色谱法测定对坐叶中木犀草素含量的方法。方法:色谱柱为Diamonsil C18(250mm×4.6mm,5μm),流动相为甲醇-0.04%磷酸溶液(60∶40),流速为1mL.min-1,柱温为35℃,检测波长为350nm。结果:木犀草素进样量在101.6~812.8ng范围内与峰面积积分值呈良好线性关系(r=0.9999);平均回收率为97.32%,RSD=0.9%(n=9)。结论:本方法简便、准确、分离效果好,可用于对坐叶的质量控制。 OBJECTIVE: To determine the content of luteolin in Hedyotis uncinella by HPLC. METHODS: The chromatographic separation was performed on Diamonsil C18(250 mm ×4.6 mm, 5 μm) with column temperature kept at 35 ℃. The mobile phase consisted of acetonitrile - 0.04% phosphoric acid(60 : 40) at a flow rate of 1 mL · min^-1; the detection wave- length was set at 350 nm. RESULTS: The linear range for luteolin was 101.6-812.8 ng (r = 0.999 9) with an average recovery of 97.32% (RSD = 0.9%, n = 9).CONCLUSION: The method is simple and accurate with good results in sample separation, and it is applicable for the quality control of H.uncinella.
