摘要
目的:观察不同浓度雌激素对卵巢癌细胞系乳腺癌/卵巢癌易感基因(BRCAI)蛋白表达的影响。方法:采用免疫组化及计算机图象分析方法,检测体外不同浓度(10-9mol/L^10-6mol/L.)的17-β雌二醇(F2)对卵巢癌OVCAR3和SKOV3细胞系BRCA1蛋白表达的影响。结果:OVCAB3和SKOV3细胞在E2的作用下,BRCA1的表达明显增加,且呈现正性剂量效应关系。结论:雌激素能够诱导卵巢癌OVCAB3和SKOV3细胞系BRCA1蛋白的表达,可通过该方法探索治疗卵巢癌的新途径。
To investigate the regulatory effect of the expression of breast cancer /ovarian cancer susceptibility gene (BRCA1 ) protein by different conentration of estrogen on ovarian caner lines. Method:The regulation effect of 17 - β estradiol (E2) on BRCA1 protein expression at OVCAR3 and SKOV3 celllines was investigated by immunohistochemistry assay and computer image analysis- R esults: under theeffect of estrogen expression of BRCA1 protein was raised which showed a dose - depedent responses ,Conclusion: Different estrogen conentration may affecte expression of BRCA1 protein in OVCAR3 andSKOV3 cell lines, by which a new approach for the treatment of ovarian caner could be investigated .
