CD40L在大鼠糖尿病性溃疡中的意义及与ICAM-1、TG的相关性

The Role of CD40L in the Rat Model with Diabetic Ulcer and the relationship with ICAM-land TG

目的:研究白细胞表面分化抗原40配体(CD40L)在大鼠糖尿病性溃疡发病中的作用及其与细胞间黏附分子-1(ICAM-1)、甘油三酯(TG)的相关性。方法:雄性SD大鼠腹腔注射中等剂量链脲佐菌素(STZ),同时给予高脂饮食建立糖尿病模型;8w末,所有大鼠下肢涂抹冰醋酸建立糖尿病下肢溃疡模型,观察溃疡变化;12w末处死大鼠,测体重并取血,测空腹血糖、放免法测空腹胰岛素、TG,酶联免疫法测sCD40L及ICAM-1,免疫组化法测皮肤组织中的CD40L。结果:注射STZ后糖尿病大鼠出现明显多饮、多尿、体重减轻症状,胰岛素水平明显下降;糖尿病并溃疡组血TG、sCD40L及ICAM-1明显高于其他组,皮肤组织中CD40L亦高于其他组;而正常对照组与溃疡对照组各项指标差异无统计学意义;sCD40L与ICAM-1之间呈显著正相关,而TG与两者之间无相关性。结论:CD40L、ICAM-1、TG可能在糖尿病足溃疡的发生中起了重要作用。

Objective： Study the role of CI40 ligand （CD40L） ,intercellular adhesion molecule-1 （ICAM-1）and Triglyceride（TG） inthe diabetic rat with lower extremity ulcer. Methods：SD male rats were injected medium- dose streptozotocin （STZ） and at the same time given high-fat diet. At the 8th weekends, all rats＇lower extremity apply acetic acid to estabish model of diabetic leg ulcers,then observe the ulcers. All rats were killed at the12th weekends, measure weight, fasting blood glucose, TG, fasting insulin by radio-immunity, sCD40L and ICAM-1 by euzymelinked immunosorbent assay （ELISA）, skin CIMOL by immunohistochemisty. Results：Diabetic rats has significant syndrome of polydipsia, polyuria and weight loss;TG, sCIMOL,ICAM-land CD40L significantly higher in the diabetes with ulcers group than other groups, while the control group and the control group with ulcers has no differents, sCD40L and ICAM-1 was obviously correlation ,TG has no obviously corre- lation with sCD40L and ICAM-1. Condusion：CD40L,ICAM-1 ,TG may play important roles in diabetic foot ulcers.
