积雪草颗粒对TGF-β1诱导的肾小管上皮细胞BMP-7表达的影响 被引量:26

摘要 目的观察积雪草颗粒(CAG)对转化生长因子β1(TGF-β1)诱导的肾小管上皮细胞NRK52E骨形态发生蛋白-7(BMP-7)及TGF-β1表达的影响。方法将体外培养的NRK-52E分为正常对照组(N组)、TGF-β1刺激组(T组)和CAG小(C1组)、中(C2组)、大(C3组)及蒙诺组(M组)。细胞培养48 h后取出,采用RT-PCR技术和细胞免疫化学技术检测其BMP-7、TGF-β1 mRNA及蛋白。结果与N组比较,T组BMP-7 mRNA及蛋白的表达明显减少,TGF-β1 mRNA及蛋白的表达均明显增加(P<0.05);C2、C3、M组TGF-β1 mRNA及蛋白表达水平均显著低于T组(P均<0.05),BMP-7 mRNA及蛋白表达水平均显著高于T组(P均<0.05);G3、M组BMP-7 mRNA及蛋白和TGF-β1 mRNA及蛋白表达水平与N组比较,P均>0.05。结论CAG可通过直接抑制NRK52E中TGF-β1的表达并维持BMP-7的表达,而发挥抗肾间质纤维化的作用。 Objective To observe the effects of centella asiatica granula(CAG) on the expression of BMP-7 and TGF-β1 of tubular epithelial ceils triggered by TGF-β1. Methods NRK52E was divided into normal control group( group N), treated with TGF-β1 ( 10 ng/ml) group ( group T) and low( group C1 ), medium ( group C2 ), high ( group C3 ) CAG group, fosinopril group( group M). The BMP-7 and TGF-β1 mRNA and protein expression of NRK52E were assessed by RT-PCR and immunohistochemistry independently. Results Compared with group N, the expression of TGF-β1 mRNA and protein markedly increased in group T( P 〈 0.05 ) ; Compared with group T, the group C2, C3 and M suppresses the high expression of TGF-β1 in NRK52E cells induced by TGF-β1 (P 〈 0.05 ) , but the expression of BMP-7 mRNA and protein was increased ( P 〈 0.05 ) ; There is no significant differences anaong the measured indexes in the group C3 and group M compared with group N ( P 〉 0.05 ). Conclusion CAG may prevent renal interstitial fibrosis through a mechanism of inhibiting the expression of TGF-β1 directly and maintain the expression of BMP-7.
