摘要
目的制备纳米级磁性多柔比星脂质体(DOX-MLP)。方法用反相蒸发法制备纳米级DOX-MLP,以正交实验设计确定最佳制备工艺。电镜观察脂质体形态,zetapal粒度分析仪测量其粒径,紫外sephadex G-50法测定其包封率,磁测仪测量其磁响应性。结果DOX-MLP呈圆形或椭圆形,粒径为(293.4±20.6)nm,包封率为30.8%±3.27%,磁响应性(50.28±7.8)emu。结论用反相蒸发法制备的磁性多柔比星脂质体平均粒径小,磁响应性高,达到纳米级标准。
Objective To prepare magnetic doxorubicin (DOX) nanoliposomes. Methods Magnetic DOX liposomes (DOX-MLP) were prepared by the reverse-phase evaporation method,an optimal technique was chosen with orthogonal design, and its diameter distribution was observed under electronic microscope. Its encapsulation efficiency was detected by the sephadex column-ultraviolet method ,the magnetic properties of the DOX-MLP were measured with a magnetometer. Result The DOX-MLP were globular or elliptic, the average particle size was (293.4 + 20.6) nm, its encapsulation efficiency was 30.8% + 3.27%, and its average magnetic response was (50.28 + 7.8) emu. Conclusion The DOX-MLP prepared by thereverse-phase evaporation method have litter diameter and high magnetic response.
