白色念珠菌细胞壁不溶性β-葡聚糖(β-glucan)抗肿瘤细胞的实验研究

摘要 目的本研究旨在观察白色念珠菌细胞壁不溶性β-葡聚糖(Cand ida A lb icans Insolub le-βG lucan,CAIBG)对XW-05肺腺癌细胞的体外生长抑制作用并初步探讨其机制。方法自制CAIBG,采用改良MTT法检测CAIBG对XW-05肺腺癌细胞的生长抑制作用,并与顺铂对XW-05肺腺癌细胞生长抑制作用相比较,再结合流式细胞仪检测加入CAIBG后肿瘤细胞的凋亡率和细胞周期分布时相。结果CAIBG对XW-05肺腺癌细胞有体外生长抑制作用,生长抑制率最高为61.27%,IC50约为0.62μg/μl;CAIBG和顺铂对XW-05肺腺癌的体外抑制作用比较,差异有显著性(p<0.05)。流式细胞仪检测结果显示CAIBG对XW-05肺腺癌细胞的细胞周期没有影响。结论CAIBG具有体外直接抑制XW-05肺腺癌细胞生长的作用。CAIBG是具有良好应用开发前景的抗肿瘤药物。 Objective In this study, we explored whether CAIBG could suppress the tumor cells growth in vitro and to explore its mech- anism. Methods Insoluble β-glucan was extracted from Candida albieans, and the reforming MTT assay was used to evaluate the growth inhibition effect of CAIBG on pulmonary adenoearcinoma XW - 05 cells in vitro. Different concentrations of CAIBG were added to cultured tumor cells in vitro at different times. The cell cycle distribution and apoptosis rate of pulmonary adenocareinoma XW-05 cells were detected by flow cytometry. Results CAIBG had inhibitory effects to the pulmonary adenoearcinoma XW-05 cells in vitro, and the highest growth inhibitory rate of the pulmonary adenocareinoma was 61.97% , and ICs0 was about O. 62 μg/μL The cancer cells' growth inhibitory effects of CAIBG and the Cisplatin had significant distinction (p 〈 0. 05). The cell cycle distribution of pulmonary adenoeareinoma XW-05 cells had no marked change after being treated with CAIBG. Conclusions CAIBG had growth inhibitory effect on pulmonary adenocarcinoma XW-05 cells. CAIBG is worthy of further research and development.
