目的分析鼻息肉患者息肉组织及其外周血的淋巴细胞免疫表型,探讨鼻息肉组织是否存在局部黏膜免疫系统。方法对17例鼻息肉患者的外周血、息肉组织及11例健康对照者的外周血,采用流式细胞术检测淋巴细胞亚群、干扰素γ(interferon-gamma,IFN-γ)和白细胞介素10(interleukin-10,IL-10)的表达水平,并进行统计学分析。结果鼻息肉患者息肉组织的CD4+T细胞(24.01%±9.20%)低于其外周血(39.61%±7.41%,P<0.01),CD8+T细胞(35.81%±6.79%)高于其外周血(27.23%±4.30%,P<0.01),CD16+CD56+细胞、CD3+T细胞在各组间差异无统计学意义(P>0.05)。鼻息肉组织T淋巴细胞分泌的IFN-γ和IL-10表达水平均高于外周血(P<0.01)。结论鼻息肉患者息肉组织及外周血T淋巴细胞亚群及细胞因子表达水平存在差异,提示鼻息肉组织可能存在局部黏膜免疫系统。
Objective To evaluate the lymphocyte subpopulations both in nasal polyps and the peripheral blood in patients with nasal polyps, and to investigate whether there was a local mueosal immune system in nasal polyps. Methods The lymphocyte subpopulations and their production of cytokines such as interferon-gamma(IFN-γ) and inter- leukin-10(IL-10) was evaluated by flow cytometry in nasal polyps and the peripheral blood from 17 nasal polyps patients and peripheral blood from 11 healthy persons. Results There were fewer CD4^+ T lymphocytes (24. 01% ± 9.20% ) and more CD8^+ T lymphocytes (35.81% ±6.79% ) in nasal polyps than in the corresponding peripheral blood (CD4^+: 39.69% ± 7.41% ; CD8^+:27.23% ± 4.30% ,P 〈 0. 01 ). No significant difference was observed in CD16^+ CD56^+ cells and CD3^+ cells among the three groups (P 〉 0.05 ). The expression level of T lymphocyte producing IFN-γ and IL-10 in nasal polyps was higher than in the peripheral blood( P 〈 0.01 ). Conclusions The difference in lymphocytes subpopulations and their production of cytokines between nasal polyps and peripheral blood might suggest that a local mucosal immune system be present in the nasal polyps.
