摘要 目的评价转染报告基因LacZ的ROSA β geo26小鼠能否作为神经干细胞(NSCs)移植的细胞来源。方法应用X-Gal结合免疫组化双标染色研究该品系小鼠NSCs植入成年大鼠纹状体内β-gal的表达。结果移植2周后在大鼠纹状体内可见表达β-gal阳性细胞,部分移植细胞向嘴侧迁移至宿主基底前脑,并分化为成熟神经元,但未观察到表达β-gal的星形胶质细胞。结论LacZ的ROSA β geo26小鼠海马的NSCs移植后能稳定表达β-gal,可作为神经干细胞移植的细胞来源。 Objective To evaluate whether LacZ transgenic ROSA β geo26 mice may be a cellular source for neural stem cell (NSC) transplantation. Methods X-Gal histochemistry combined with immunohistochemistry doublestaining was performed to localize the expression of β -galactosidase (β-gal) in NSCs derived from the hippocampus of newborn ROSA β geo26 mice after engraftment into adult rat striatum. Results β -gal positive cells were observed in the rat striatum 2 weeks after the transplantion. A proportion of engrafted cells migrated rostrally, reached the host basal forebrain and differentiated into mature neurons, but no β -gal positive astrocyte was observed. Conclusion NSCs derived from the hippocampus of LacZ transgenic ROSA β geo26 mice could expressβ -gal stablely, be a cellular source for neural stem cell transplantation.
