小鼠精原细胞染色体畸变分析中几个影响因素的探索

摘要 本文对小鼠睾丸精原细胞染色体畸变分析方法中的低渗液种类、低渗时间、染液浓度等几个重要的影响因素进行了试验比较,试验程序主要参照《中华人民共和国农药评价标准》(GB15193.8-94),试验结果表明:75mmol/L氯化钾低渗液化于10g/L柠檬酸钠,在37℃条件下,低渗时间以10~15分钟为佳;pH6.8的1∶6Giemsa染液(7.6g/L)的染色效果较pH6.8的1∶1Giemsa染液的染色效果好。在对农药阔草净进行小鼠睾丸精原细胞染色体畸变分析时发现:阔草净1000mg/kg,5000mg/kg,10000mg/kg三个剂量组精原细胞染色体畸变细胞率分别为0.8%、1.2%和1.2%,与阴性对照的染色体畸变细胞率(0.9%)比较无显著性差异(P>0.05),说明在本试验条件下,农药阔草净对小鼠睾丸生殖细胞染色体无损伤作用。 This paper studied main affecting factors in spermatogonium chromosome aberration test of mice. They are kinds of low osmosis liquors, the time of low osmosis, the concentration of Giemsa staining solution. This test according to the standard method of assessment toxicity of pesticide. The results showed 75mmol/L KCL is better than 10g/L sodium citrate acid as low osmosis liquor. 10-15 minutes are suitable time of low osmosis when the temperature is 37℃, 1∶6 Giemsa staining solution(7.6g/L) is better than 1∶10 proportion when pH value is 6. 8. Then we studied a pesticide named Kuocaojing, the results showed: when the dosages of Kuocaojing are 1000mg/kg、 5000mg/kg、10000mg/kg, the rate of chromosome aberration are 0.8%、1.2% and 1.2%. Comparing them to control group(0.9%), the results have no significant difference(P>0.05).
