摘要
利用新色原建立测定血清葡萄糖浓度的方法.以葡萄糖氧化酶为基础用双试剂结合速率法进行定量分析.结果:葡萄糖浓度达50.0mmol/L反应仍呈良好的线性;回收率为99.9%,日内CV为0.88%,日间CV为2.03%,除高浓度的血红蛋白外,能有效地避免血清中氧化物、浊度、色素等物质的干扰,测定结果与葡萄糖氧化酶/酚法和葡萄糖激酶法有良好的相关性,参考值范围为3.88~5.56(4.72±0.43)mmol/L.结论:该方法由于试剂稳定,成本低,测定方法可靠,适合在具有双试剂分析功能的自动生化分析仪上使用.
This paper describes a double-reagent colorimetric assay for glucose determination in human serum using a new chromgen. Serum glucose will be determed by glucose oxidase with double-reagents and kinetics method. Glucose concentrations in serum up to 50 mmol/L can be measured. The average analytical recovery of glucose in diffierent human sera was 99. 9%. within day CV was 0. 88% and between day CV was 2. 03%. The effect of interfering serum reducing compounds does not seem to be large, even with severafold increased amounts of the compounds tested. An exception to this is hemoglobin at very great concentrations. The results of the recommrnded procedure correlated well with those of the phenol/4 - aminoantipyrine method of Trinder and metbod of glucose kinase. The method is stability, reliability, economical acceptability,and can be used to biochmical automatic analyzer witb double-reagents, therefore can be recommended for routine hospital assay of serum glucose.
