摘要
采用MTT法和流式细胞术,对粘细菌Stigmatella WXNXJ-B次级代谢产物生物活性进行了评价,并对其作用机制进行了研究。结果表明,Stigmatella WXNXJ-B产生的活性物质主要为内酯类物质,CF100部分对小鼠黑色素肿瘤B16细胞抑制能力最强,作用时间48h和72h,其IC50分别为9.94mg/L和8.64mg/L;当质量浓度达到20mg/L,48h后凋亡细胞表面出泡,凋亡小体形成,G0/G1期的细胞由80.9%下降至24.2%,S期和G2/M期的细胞由15.4%增加到53.6%和3.8%增加到22.2%,从而使细胞凋亡。
The bioactivity of secondary metabolites produced by myxobacteria Stigmatella WXNXJ - B was evaluated and the action mechanism was investigated by using the MTF method and the flow cytometry. The results showed that the bioactive components containing the lactones by the qualitative analysis experiment. The subfraction CF100 had a higher anti-tumor activity on the mouse melanoma B16 cell line than other subfraetion after 48 h and 72 h treatment. Its IC50 were 9.94 mg/L and 8.64 mg/L respectively. The cell treated with CF100 at the concentration of 20 mg/L for 48 h began to form blebs and apoptotic bodies. The cell apoptosis happened because CF100 caused G0/G1 phase attenuation from 80. 9% to 24. 2% and the cells were accumulated in S and G2/M phases from 15.4% to 53.6% and from 3.8% to 22.2% respectively.
