摘要
目的研究B71基因转染小鼠EL4淋巴瘤细胞的致瘤性,为制备基因转染瘤苗提供实验依据。方法将B71基因转染小鼠EL4淋巴瘤细胞后,用G418选择和克隆的方法获得高表达B71分子的EL4B71+细胞,然后进行小鼠的体内致瘤试验。结果EL4B71+细胞接种小鼠后,与野生型EL4(EL4Wt)相比,肿瘤结节形成时间和荷瘤小鼠存活时间明显延长,EL4B71+细胞协同IL2接种小鼠后,效果更加显著。结论B71基因转染小鼠EL4淋巴瘤细胞的致瘤性比野生型EL4(EL4Wt)低。
Objective To study the Oncogenicity of rat EL4 lymphoma cell transfected with Gene B71 and to provide experimental basis for preparing tumor vaccine by transfected gene.Method oncogenic experiment in vivo was carried out after EL4B71+ cell,which was the high expressioon of B71 molecule,was obtained by G418 after rat EL4 lymphoma cell being transfected with gene B71.Result Compared with wild type EL4(EL4Wt),the formation time of tumor nodes and survival time of rats with tumor were remarkably prolonged.The result was more significant when EL4B71+ and IL2 were used together. Conclusion The oncogenicity of rat EL4 lymphoma cell transfected with gene B71 is lower than that of wild type EL4 lymphoma cell
