摘要
建立了高效液相色谱(HPLC)测定小儿复方氨基酸注射液中各种氨基酸含量的方法。采用Kromasil C18(250×4.6mm,5μm)色谱柱,以2,4-二硝基氟苯为衍生试剂,梯度洗脱。流动相A为乙腈:水(50:50,V/V),流动相B为0.04mol/L的磷酸二氢钾溶液(pH=6.8);流速为1.0mL/min;检测波长为360nm。所测氨基酸的线性范围为0.008-0.2213mg/mL,平均回收率在97.85%~102.86%之间。
A pre-column derivatization high performance liquid chromatogrphy method for the determination of amino acids in the children compound amino acid injection was established. The sample was derivatized with 2,4-dinitrofluorobenzene(DNFB). Kromasil C18 column (250 × 4.6 mm, 5μm) was used. The gradient elution was performed using mixed solution of acetonitrile and water (50:50,V/V), and 0.04 mol/L KH2PO4 (pH=6.8) as the mobile phases respectively at a flow rate of 1.0 mL/min. All the amino acids measured show good linear relationship within 0. 008 - 0. 2213 mg/mL and average recoveries between 97.85% and 102.86%.
