摘要
目的探讨食管癌组织中9p21(D9S171)杂合性丢失在食管癌发生中的作用。方法抽提正常和肿瘤组织中的DNA,应用9p染色体微卫星标志,经聚合酶链反应,检测食管癌组织中9p21的杂合性丢失。结果32例食管癌组织,14例出现9p21位点条带的缺失,杂合性丢失率为43.7%。杂合性丢失与临床病理之间无明显关系。结论杂合性丢失在高发区食管癌病人中有较高的检出率,表明杂合性缺失与食管癌的形成密切相关。
Objective Our purpose was to determine the incidence of loss of heterozygosity(LOH)on 9p21 in human esophageal cancer and its correlation with esophageal cancer.Method Genomic DNA were isolated from 32 cases of paired normal and tumor esophageal tissues, and loss of heterozygosity (LOH) on 9p21 (D9S171 loci) were analyzed by using highly polymorphic microsatellite marker and polymerase chain reaction.Result Of there 32 patients with esophageal cancers, loss of heterozygosity on 9p21 (D9S171 loci) was found in the tumor tissue of 14 cases, its rate was 43.7%.There were not statistically significant correlations between LOH and clinical pathology.Conclusion The highest frequency of LOH suggests that LOH is an event involved in esophageal cancer progression.
