摘要
目的：研究人胞苷转移酶（ＣＴ）ｃＤＮＡ在鼠巨噬细胞中的表达。方法：按照Ｔ．Ｍａｎｉａｔｉｓ等人的方法人转染的巨噬细胞中提取总ＲＮＡ，电泳分离ＲＮＡ，转移ＲＮＡ至尼龙膜并固定，制备３２ｐ标记的胞苷转移酶ＤＮＡ探针，杂交及放射性自显影。结果：对照组无表达，同类转染的细胞均有不同程度的表达。结论：制备探针前应用电泳法确定ＣＴｃＤＮＡ片段的浓度比紫外光吸收法更准确适用。
objective:To study the expreasion of humen CT cDNA in the mouse macrophages. Method:Using the CT cDNA(1200bp)which was gotten from plasmid vector,hybridization and autoradiograph.We Prepared the RNA using a reagent from Biotcx;Running an agarose gel to isolate RNA and tranafered and fixeed RNA to nylomembren;Labeling probe for the CT mRNA Results:there was difference in expression between the control and specimen.Concinsion:it is easy and exact to check the quantity of CT cDNA.QQuick spin colum is better for getting labeling probes from mixture
