摘要
目的：应用单克隆技术制备鼠抗人ＩＶ型胶原（ＩＶ－Ｃ）单克隆抗体。用氯胺－Ｔ方法标记ＩＶ－Ｃ，应用免疫竞法建立ＩＶ－Ｃ放射免疫分析方法，其检测范围２０￣５００ｕｇ／Ｌ。方法和结果：通过特异性、精密度、准确性、亲和常数等实验方法学检验，表明ＩＶ－Ｃ放射免疫分析方法符合临床应用要求。应用该方法检测３０例肝炎患者、１５例肝硬化患者、３５例ＮＩＤＤＭ患者、２５例甲亢患者；除ＮＩＤＤＭ、甲亢外。
Objective: To found a radioimmunoassay (RIA) method of Type Ⅳ Collagen(Ⅳ-C) for clinic applications.Method: Mouse monoclonal Ⅳ-C antibody were prepared by monclonal technology.Ⅳ-C were labelled the chloramine-T method.Results:Method of detection Ⅳ-C was deveolpedn by RIADection range:20-500ug/L. Conclusion: The RIA of Ⅳ-C was tested by sensitivity,affinity constant,sensitivity,precision and the results indicate that RIAof Ⅳ-C fits clinic applications. Concenfrations of serum Ⅳ-Cwere measured by RIA in 30 cases hepatitis and 15 cases cirrhosis of liver and 35 cases hepatitisand 15 cases hperthyroidism and compared with those in 50cases normal humans, the concertrations of serum Ⅳ-C in all patients were markedly increased iespectivedly expect NIDDM and and and hyperthyroidism were significantly higher than normal humsns(P<0.05). These results suggest that serum Ⅳ-Ceoncentration is valuble in clinical study of liver and other relative diseases.
