摘要 用尿素抽提及DE_(32)^-纤维素离子交换柱层析,从牛大脑白质中提取GFAP,经鉴定具理化性质符合文献报道。用提取的GFAP免疫BALB/c小鼠,经杂交瘤技术,获得稳定分泌抗GFAP单克隆抗体的杂交瘤细胞株AG_1,染色体众数为68,属小鼠IgG_1亚类,免疫印迹试验时仅与GFAP起特异性反应,ABC免疫组化显示AG_1单抗与星形胶质细胞及其肿瘤呈阳性反应,而与非胶质细胞及其肿瘤呈阴性反应。 Glial fibrillary acidic protein (GFAP) was isolated and purified from bovine brain white matter by urea extraction and DEAE-cellulose anion-exchange chromatography. The extracted GFAP was characterized and its physicochemical properties are similar to those reported. Mouse hybridomas producing monoclonal antibodies(McAb)to GFAP (AG1) have been cloned and established. Its mean chromosome unmber is 68.The McAb was shown to be of the mouse IgGl subclass. It reacts specifically with GFAP in immunoblotting.The tissue specificity of this McAb was tested on normal and tumor tissues with immunohisto-chemical techniques. AG1 showed positive reactions with astroglia and its tumors. On the other hand, the non astroglia and its tumors showed negative reactions.
