摘要
目的:探讨非甾体类消炎镇痛药舒林酸对卵巢癌裸鼠腹腔移植瘤生长的影响。方法:将卵巢癌细胞SKOV3注射于裸鼠腹腔建立卵巢癌移植瘤模型,在成瘤之前和成瘤之后两个时间段分别采用舒林酸灌胃,计算各组抑瘤率,采用RT-PCR、Western blot观察MMP-2、VEGF的表达变化。结果:舒林酸预防组对裸鼠卵巢癌生长有明显抑制作用。舒林酸预防组在mRNA和蛋白水平表达MMP-2和VEGF均显著降低。结论:早期应用舒林酸能够抑制裸鼠卵巢癌腹腔移植瘤的生长,其机制可能与降低VEGF、MMP-2表达有关。
Objective: To explore the effect of sulindac on growth of transplantable ovarian carcinoma of nude mice model. Methods : Nude mice model of transplantable ovarian carcinoma was established by injecting ovarian carcinoma cell SKOV3 into abdominal cavity, then sulindac was used to lavage before and after tumorigenesis, the suppressive rate of carcinoma was calculated, and the expression levels of MMP - 2 and VEGF were detected by RT - PCR and Western blot. Results : Sulindae inhibited the growth of transplantable ovarian carcinoma significantly. In sulindac prevention group, the levels of VEGF mRNA and protein, MMP - 2 mRNA and protein decreased significantly. Conclusion: Early administration of sulindac can inhibit the growth of transplantable ovarian carcinoma signifieantly, which maybe related to downregulation of MMP- 2 and VEGF.
