槲皮素干预实验性慢性肾衰大鼠肾脏SOD与LPO变化 被引量:4

摘要 目的:观察槲皮素防治动物实验性慢性肾衰的效果并初步探讨作用机理。方法:给实验大鼠饲以含腺嘌呤饲料2月造成慢性肾衰;干预组经口给予槲皮素(100 mg/Kg.d×42 d)治疗;按试剂盒指定方法测定SOD及LPO;测定羟脯氨酸以观察肾组织纤维化程度。结果:与肾衰对照组比较,干预组大鼠肾脏SOD显著升高(66.5 u/mL±34.9 u/mL或7.68 Ku±2.98 Ku/肾vs27.5 u/mL±24.3 u/mL或3.26Ku±2.63 Ku),LPO亦显著增加(8.9 nmol/mL±3.0 nmol/mL或103.3 nmol/mL±16.1 nmol/肾vs 2.9nmol/mL±1.2 nmol/mL或39.2 nmol/mL±17.5 nmol/mL),肾组织纤维化显著减轻(平均每肾羟脯氨酸含量735.5μg±377.7μg vs1540.3μg±358.9μg),肾组织细胞NF-κB P65丢失情况明显减轻。结论:槲皮素对大鼠实验性慢性肾衰具有显著防治作用。其作用可能与保护肾脏细胞及抗氧化作用,减轻细胞损伤有关。 objective: The prevention and curing effects of Quercetin in rats with experimental chronic renal failure were observed and its mechanism was primarily studied. Methods: The rats in control and treating groups were fed with 0.75% adenine in feed for 2 month to be build up of chronic renal failure; and the quercetin were administered orally ( 100 mg/Kg/d) for 42 days to the rats in treating group, beginning at the 15th day. The superoxide dismutase (SOD) and lipid peroxides (LPO) were determined with the methods designed in the bought kits of assaying agents. The renal tissue sclerosis was assayed by determining its hydroxyproline (Hyp) . The NF-KB in the renal cells were assayed with a immunoehemical assay. Results: The renal SOD in treated group were increased significantly when compared with the control group (66.5 u/mL ±34. 9 u/mL or 7.68 Ku ±2. 98Ku/Kedney vs 27.5 u/mL±24. 3 u/mL or 3. 26 Ku ± 2. 63 Ku), ( P 〈 0. 01 or P 〈 0. 001 ) ; the renal LPO were increased ether ( 8. 9 nmol/mL ± 3. 0 nmol/mL or 103. 3 nmol/mL ± 16. 1 nmol/Kedney vs 2. 9 nmol/mL ± 1.2 nmol/mL or 39. 2 nmol/mL ± 17. 5 nmol/mL) ; the renal tissue sclerosis changes were reduced ( mean quantity of Hyp per kidney, 735.5 ± 377.7ug vs 1540. 3 ± 358. 9ug) , and the losses of NF-κB P65 in the renal cells were less serious. Conclusion: The results suggested that qereetin is of prevention and curing effects on chronic renal failure in rats ; the effects may be related to the mechanisms of its preserving renal cells and anti-oxidation, preventing and remeding injuries in cells of the kedney of treated rats.
