改良MTT法检测胃癌乳腺癌体外药物敏感性

In vitro sensitivity testing of fresh human stomach cancer and mammary cancer using modified MTT colorimetric assay

采用改良的MTT法对53例胃癌、乳腺癌进行体外药敏试验。结果表明，改良的MTT法，即多次贴壁分离癌组织中夹杂的成纤维细胞，且细胞悬液培养过夜后又能排出污染的标本，因此提高了体外药敏的准确性。改良的MTT法优点有：（1）获得了较纯化的癌细胞；（2）培养过夜能使受损伤的细胞恢复活性，增加癌细胞含量，提高实验成功率。

In vitro drug sensitivity testing of human tumor cells is the basis of laboratory to treatthe individual tumor, our country is now using MTT colorimetric assay universally, but this artical carriedout in vitro drug sensitivity testing of 53 cases with stomach cancer or/and mammary cancer. The resultshowed that modified MTT colorimetric assay method can cancel fibroblast in tumor organization and itscell suspention cultured through night can excule contaminated samples, so it can promote accuracy of invitro drug sensitlvity testing. This method has merit: (1) obtaining purer cancer cells ; (2) cell suspentioncultured through night can recover cell activity in damaged and add quantities of cancer cell and promotethe rate of snccess in laboratory.
