摘要
基于中蜂囊状幼虫病病毒基因组序列AF469603中的RNA依赖的RNA聚合酶基因,建立该病毒的半套式RT-PCR检测方法。利用建立的方法检测有典型临床症状的病死中蜂幼虫,可扩增到特异的目的片段,序列分析得出,PCR扩增片段与AF469603同源性高于97%,与其他病原无同源性,表明该方法可用于临床诊断和流行病学研究。
A specific semi-nested RT-PCR based on the RNA-dependent RNA polymerase gene in AF469603 of Chinese bee (Apis sinensis) sacbrood virus was established to detect Chinese bee sacbrood virus. The method was applied to detect clinically dead larval samples. Results showed that the specific fragment was gained from the samples. Sequence homology between the amplified segment and that of AF469603 was as high as 97% and no sequence of other pathogens was amplified. In conclusion, the RT PCR method repre- sents a powerful tool for diagnosis of Chinese bee sacbrood virus infection and epidemiological investigation.
