淀粉载体固定化脲酶的研究 被引量:2

摘要 脲酶是一种来源广泛的植物蛋白酶,能高效、专一的催化尿素分解。本文利用双醛淀粉(DAS)作为酶固定化载体对脲酶进行固定化。同时研究反应温度,反应时间,pH,载体的醛基含量,载体与酶的配比对固定化酶活性的影响。经实验确定,最佳工艺条件为:双醛淀粉载体醛基含量80%,反应pH值6.0,反应温度25℃,反应时间25h,酶与载体的配比为1:1(mL/g) As one kind of origin widespread plant protemase, the urease can selectively catalytic decomposition of urea with high effect. In this paper, dialdehyde starches were used as carrier for immobilization of urease. The effects of temperature, time, pH, aldehyde group content of carriers and carrier/enzyme ratio on immobilizing urease were studied. The best conditions were determined as follows: 80% of aldehyde in dialdehyde starch, 6.0 of the reaction pH value, 25℃ of reaction temperature, 25h of reaction time, and the enzyme-cartier ratio of 1:1 (mL / g)
