摘要
以胶原蛋白和透明质酸为基材,用碳化二亚胺交联合成可用于药物的控制释放的医用可降解水凝胶。考察了不同胶原蛋白与透明质酸配比对水凝胶吸水率和表面形状的影响,采用生理盐水、磷酸盐缓冲液、I型胶原蛋白酶和透明质酸酶评价了这种水凝胶的生物降解性,并对载药水凝胶进行了体外降解释药实验。研究表明:胶原蛋白与透明质酸的配比为1.5:2时得到的复合水凝胶吸水率和表面形状等因素都较好;水凝胶在混合酶溶液中降解较为迅速;布洛芬(IBF)采用包埋方式载药的药物释放速率比浸泡方式的速率慢。
The degradable medical hydrogel was synthesized for controlled drug releasing using collagen and hyaluronic acid as substrate and carbodiimide as cross-linking agent. The effects of ratio of collagen to hyaluronic acid on the water absorption rate and surface shape was studied, and biodegradability of the hydrogel was evaluated by determining the normal saline ,phosphate buffer solution, collagenase type I and hyaluronidase. The drug-release performance of the drug-loading hydrogel was investigated in vitro. The results indicated that the compound hydrogel showed better water absorption and staface shape when the ratio of collagen to hyaluronic acid was of 1.5:2. The hydrogel can degrade quickly in an osmotic solution containing an enzyme mixture. The drug-release rate of drug IBF with embedding method was slower than that with the immersing technique.
