摘要
探讨了葡聚糖酶产生菌的初筛培养基组成。通过单因素试验,确定了培养基中碳源的种类、用量及刚果红的用量和使用方法等对初筛效果的影响。结果得到优化后的初筛培养基配方为:葡聚糖0.5%,NaNO3 0.2%,K2HPO4 0.1%,KCl 0.05%,MgSO4 0.05%,FeSO40.001%,刚果红0.005%,琼脂2%。此初筛培养基葡聚糖和刚果红的用量少,形成的透明圈大,能够达到比较好的初筛效果。
A culture medium for primary screening of medium for glucanase producing strains was studied. The kinds and amounts of carbon source and the using methods and amounts of congo red on the culture of the strains were tested. Single factor experiment showed that the optimal medium consisted of 0.5% glucan, 0.2% NaNO3, 0.1% K2HPO4, 0.05% KCl, 0.05% MgSO4, 0.001% FeSO4, 0.005% congo red and 2% agar. Using this media, good primary screening effect was achieved with low glucan and congo red consumption and large transparent circle.
