摘要
利用扩增长度多态性(AFLP)分子标记技术,构建湖南省现行推广的10个桑树品种的指纹图谱,作为该省区桑树遗传育种辅助选择和品种鉴定的重要依据。从36对AFLP引物中筛选出带型丰富、多态性较高的5对引物对10份桑树品种材料进行扩增,共检测到344条多态性条带,多态性比率达27.88%。品种之间的遗传相似系数以及UPGMA聚类分析结果与基于形态学方面的分类结果一致,在遗传相似系数0.79处,10个桑树品种聚为3类:三倍体品种湘桑6号单独聚为一类,其余9个品种聚为一类,其中广东桑品种苗33又单独聚为一类,8个鲁桑品种聚为一类。
Amplified fragment length polymorphism(AFLP) molecular marker technology was employed to construct the fingerprints for 10 current mulberry cultivars in Hunan Province.The obtained fingerprints can be used as important basis for assisted selection of mulberry breeding and variety identification in the province.Among 36 primer pairs used,5 were found to yield rich band patterns and high polymorphism.After amplification with these 5 primer pairs to 10 mulberry cultivars,altogether 344 polymorphic DNA bands were detected.The polymorphism rate reached 27.88%.The coefficient of genetic similarity between cultivars and UPGMA cluster analysis were consistent with the classification based on morphological observations.The coefficient of genetic similarity was 0.79 and the 10 mulberry cultivars could be clustered into 3 groups.The triploid cultivar Xiangsang 6 formed a single cluster and the rest 9 cultivars formed another cluster,among which Miao 33,a cultivar of Guangdong Mulberry,formed a separate clade and the rest 8 cultivars of Lu Mulberry formed another cluster.
