摘要
目的研究ZnPPⅨ对人胃癌腹膜细胞GC9811-P凋亡的影响。方法采用MTT法测定GC9811-P细胞在ZnPPⅨ作用48 h后生长抑制情况;流式细胞仪观察ZnPPⅨ对GC9811-P细胞周期和细胞凋亡的影响;western blot免疫印迹法观察ZnPPⅨ对细胞系CIAPIN1、Bcl-2、Bax蛋白表达水平的影响。结果各浓度值的ZnPPⅨ均可抑制GC9811-P细胞增殖,并促进肿瘤细胞凋亡,并降低CIAPIN1和Bcl-2/Bax蛋白的表达水平。结论ZnPPⅨ可诱导GC9811-P细胞的凋亡,其作用机制可能通过调控CIAPIN1基因影响Bcl-2/Bax分子的表达。
Objective To explore the effect of ZnPPⅨ on the apoptosis of human gastric cancer cell line GC9811-P with peritoneal metastasis. Methods After GC9811-P cells were treated with ZnPPⅨ for 48 hours, cell proliferation was detected using MTT assay. Flow cytometry was used to detect the apoptosis of GC9811-P cells, and western blot was used to check the expression of CIAPIN1, Bel-2/Bax. Results Zn- PPⅨ inhibited the proliferation of GC9811-P cells at the concentrations of 1 × 10^-2 to 1 × 10^-9 mol/L, and cell arrested in G1/S phase. The proportions of apoptosis cells were significantly increased compared with controls. The expression of CIAPIN1 and Bcl-2/Bax was decreased after incubated for 3h with 1 × 10^-5 mol/ L ZnPPⅨ Conclusion ZnPPⅨ induce the apoptosis of gastric cancer cell lines GC9811-P with peritoneal metastasis by suppressing of the expression of CIAPIN1, and affection of its downstream apoptosis factors Bcl-2 and Bax.
