椒枝软胶囊对哮喘大鼠气道黏蛋白影响的实验研究 被引量:5

摘要 目的:探讨椒枝软胶囊对哮喘气道黏蛋白(Muc5ac)高分泌的影响及作用机理。方法:将健康雄性SD大鼠48只随机分为空白对照组、模型组、强的松组、椒枝软胶囊高剂量组、椒枝软胶囊中剂量组、椒枝软胶囊低剂量组,除空白对照组外,以卵蛋白氢氧化铝混悬液腹腔+多点皮下注射,卵蛋白雾化攻击制作哮喘大鼠模型,从第13天起给予相应药物5天,末次激发后48h处死大鼠,以HE染色观察肺组织病理,阿尔辛蓝-过碘酸雪夫染色观察杯状细胞,免疫组化法观察Muc5ac阳性表达情况。结果:椒枝软胶囊可减轻哮喘大鼠气道炎症反应,椒枝软胶囊高、中剂量组气道杯状细胞比值和Muc5ac阳性表达光密度值明显低于模型组(P<0.05或P<0.01)。结论:椒枝软胶囊可减少Muc5ac高分泌,其作用机制可能是通过抑制气道炎症和气道杯状细胞增生或化生。 Objective:To investigate the effect on high airway mucin secretion in asthma with jiaozhi soft capsules and its mechanism of action. Methods: The forty - eight male SD rats were randomly divided into six groups: the control group, the asthma model group, the prednisone group, the high does of Jiaozhi soft capulses group, the medial does of Jiaozhi soft capulses group, the low does of Jiaozhi soft capulses group. The other five groups SD rats except the control group were sensitized with OVA and Al(OH) 3 by intraperltoneal injection and subcutaneous injections and attacked by atomized OVA. The rats were given corresp medicine since the 13th day and executed 24 hours later after the last atomization. Observe the pathology of lung tissue with HE staining, Observe the amount of goblet cells with AB - PAS staining. Observe the positive expression of MucSac with immunohistochemisty. Results: The degree of asthma airway Inflammation was alleviated in the groups of Jiaozhi soft capsules. The proportion of airway goblet cells and the optical density value of Muc5ac positive expression in the groups of Jiaozhi soft capsules with high dose and medium dose are obviously lower than which in the asthma model group(P 〈0.05 or P 〈0.01 ). Conclusion: The Jiaozhi soft capsules can decrease MucSac secretion through inhibiting the inflammation and tbe proliferation/metaplasia of the goblet cells in asthma airway perbapsly.
