摘要
目的通过实验研究观察肾衰泻浊丸对腺嘌呤致肾间质纤维化大鼠骨成型蛋白-7(BMP-7)及受体(BMP-7RⅡ)的影响,探讨肾衰泻浊丸抗肾间质纤维化的机制。方法雄性Wistar成年大鼠68只,随机取8只正常大鼠作为空白对照组,其余采用腺嘌呤致肾间质纤维化大鼠模型,并随机分为模型组、尿毒清对照组、肾衰泻浊丸低剂量组、肾衰泻浊丸中剂量组和肾衰泻浊丸高剂量组。药物干预60d,观察实验大鼠治疗后肾间质纤维化情况,分别采用免疫组织化学法、Western Blot法、半定量RT-PCR检测肾组织中BMP-7及BMP-7RⅡ的表达情况。结果肾衰泻浊丸能够改善肾小管间质损伤,能够升高CRF大鼠肾组织中BMP-7及BMP-7RⅡ蛋白的表达,与对照组比较有显著性差异(P<0.01);能够上调肾组织中BMP-7mRNA及BMP-7RⅡmRNA的表达,与对照组比较有显著性差异(P<0.01),且呈剂量依存性。结论肾衰泻浊丸可能是通过增加BMP-7及BMP-7RⅡ的蛋白表达,从而减轻肾间质纤维化的程度,延缓肾间质纤维化的进展。
Objective To investigate the effects of "Shenshuai Xiezhuo Pill" on BMP-7 and BMP-7R Ⅱ expressions in renal interstitial fribrosis rats to illustrate its mechanism in relieving renal interstitial fibrosis. Methods In the experiments, the ronal interstitial fibrosis model was duplicated by adenine. Totally 68 adult male Wistar rats were used, among which 8 rats were taken as control group, and the others were modeled to be of renal interstitial fibrosis and then randomized into model group, control group, low-dose pill group, middle-dose pill group and high-dose pill group. After 60-day drug intervention, Masson staining method for pathologic slice was used to observe the pathlologic change of the rats' kidney tissues; the expressions of BMP-7 and BMP-7R Ⅱin kidney tissues were detected respectively by immunohisto-chemisto', Western Blot method, and senti-quantitative RT-PCR. Results "Shenshuai Xiezhuo Pill" could obviously decrease renal interstitial injury, elevate BMP-7 and BMP-7RⅡexpressions, BMP-7 mRNA and BMP-7R Ⅱ mRNA expressions, with significant differences in comparison with control group, and the elevation is dependent on the dose of "Shenshuai Xiezhuo Pill". Conclusion "Shenshuai Xiezhuo Pill" can increase BMP-7 and BMP-7R Ⅱ expressions in kidney tissues to relieve renal interstitial fibrosis and delay the progress of renal interstitial fibrosis.
