摘要 目的探讨不同强度运动对大鼠大脑皮质内血管内皮细胞生长因子(VEGF)表达的影响,为深入研究运动性中枢疲劳的发生机制提供神经生物学实验资料。方法4月龄雄性SD大鼠24只,随机分为3组,每组8只,即对照组(C组)、中等强度运动组(M组)和大强度运动组(S组)。C组大鼠不加任何干预;M组和S组大鼠分别在进行为期5周不同强度的游泳训练。C组、M组和S组大鼠5周游泳训练完成后即刻灌注取材,采用免疫组化方法检测大鼠大脑皮质血管内皮生长因子(VEGF)的表达。结果各组大鼠大脑皮质VEGF阳性表达部位定位于神经元细胞和内皮细胞胞浆。M、S组与C组比较,大鼠大脑皮质VEGF阳性表达IOD值则呈显著性增高(P<0.01);S组VEGF阳性表达显著高于M组(P<0.05)。结论不同强度运动可引起大鼠大脑皮质VEGF表达特异性增加,VEGF的表达强度与运动强度成正比。提示VEGF的增加可以促进大脑皮质血管的生成,改变血-脑屏障的通透性,而且可能对神经元细胞具有保护作用。 Objective: To explore the effect of exercises of different intensity on the expression of brain cortex VEGF to provide neurobiological experiment material for in - depth research on the occurrence mechanism of exercise - induced central nerve fatigue. Methods : 24 male SD rats of four months old are randomly divided into three groups, the C group, the M group and the S group, each group having 8 rats. Rats in the C group are left without intervening, while rats in the M and S groups are given five - week swimming exercises of different intensity. Vaseularization and selection are im- mediately done after the five - week swimming exercise, and the Immunohistochemical method is employed to test the expression of brain cortex VEGF. Results : The positive expression parts for brain cortex VEGF of rats in each group is located in neurons and endothelial cell cytoplasm. Compared with the C group, the M and S groups witness a marked increase in the IOD of brain cortex VEGF ( P 〈 0.01 ). The positive expression for VEGF in S group is markedly higher than that in the M group( P 〈 0.05). Conclusion: Exercises of different intensity can lead to increase in the expression specificity of rat brain cortex VEGF. The expression intensity of VEGF is positively correlated to the exercise intensity. Increase in VEGF can promote the growth of brain cortex blood vessel, change the transparency of blood - brain barrier and possibly protect neurons.
