苦参碱对大鼠急性心肌缺血性损伤的保护作用 被引量:7

摘要 目的研究苦参碱对大鼠急性心肌缺血性损伤的保护作用。方法将SD大鼠随机分为7组:正常对照组、假手术组、模型组、硝酸甘油组、苦参碱2.55、、10mg/kg剂量组。应用左冠状动脉前降支(LAD)结扎法制备大鼠急性心肌缺血模型。记录结扎前、结扎后1、5、15、306、0min大鼠心率(HR)、平均动脉血压(MAP);应用分光光度计法检测血清中肌酸激酶(CK)活性和一氧化氮(NO)含量。结果静脉注射苦参碱5、10mg/kg均可使大鼠HR、MAP明显升高(P<0.05或P<0.01),此作用在左冠状动脉前降支结扎后15min最明显。静脉注射苦参碱5、10mg/kg,可降低急性心肌缺血大鼠血清CK活性(P<0.05或P<0.01);静脉注射苦参碱2.5、5、10mg/kg,可增加急性心肌缺血大鼠血清NO含量(P<0.05或P<0.01)。结论静脉注射苦参碱对大鼠急性心肌缺血性损伤有保护作用。 Objective To explore myocardial protective effects of matrine(MT) on acute myocardial ischemic injury(AMI) in rats.Methods Sprague-Dawley rats were divided into seven groups,i.e.,control,sham operation,AMI,nitroglycerin+AMI,AMI+MT 2.5mg/kg,5mg/kg,or 10mg/kg.Rat models of AMI were established by ligation left anterior descending coronary artery.At pre-occlusion and 1,5,15, 30,60 min after left anterior descending artery(LAD) occlusion,heart rate(HR) and mean arterial pressure(MAP) were recorded.The activity of creatine kinase (CK) and content of nitric oxide (NO) in blood were measured with spectrometer. Results After LAD occlusion , HR, MAP were significantly increased ( P 〈 0.05 or P 〈 0.01 ) in AMI rats after MT administration at the dosage of 5 mg/kg, 10 mg/kg. At 15 min after LAD occlusion. The CK activity was significantly reduced in AMI rats after MT administration at the dosage of 5 mg/kg, 10 mg/kg (P 〈 0.01). the NO content was significantly increased in AMI rats after MT administration at the dosage of 2.5 mg/kg, 5 mg/kg, 10 mg/kg (P 〈 0.05 or P 〈 0.01) . Conclusion Intravenous matrine can protect acute myocardial ischemic injury in rats.
