氧化苦参碱干预重症急性胰腺炎的实验研究 被引量:1

摘要 目的:探讨氧化苦参碱(OM)对实验性重症性胰腺炎(SAP)的防治作用及作用机制。方法:80只SD大鼠随机分为3组:假手术(SO)组、生理盐水防治(SAP-NS)组、氧化苦参碱防治(SAP-OM)组。5%牛胆酸钠胰胆管逆行注射建立SAP大鼠模型。SAP-OM组于造模前30分钟尾静脉注入OM(30mg/100g);NS组则尾静脉注入双蒸水(0.5ml/100g)。术后3H、6H各组分批处死动物,观察各组大鼠腹水量、血清TNF-α、IL-1β等;切取胰腺组织,计算胰腺湿干重比、评定胰腺组织损伤积分、检测MPO、TNF-α mRNA、IL-1β mRNA等。结果:SAP—OM组3H、6H湿干重比率、胰腺组织病理学积分、腹水量均低于SAP-NS组(P〈0.01);SAP-OM组各时间点血清TNF-α、IL-1β均低于SAP-NS组(P〈0.01)。结论:OM可减少TNF-α、IL-1β等炎症因子的分泌,从而对SAP具有防治作用。 Objective: To investigate the therapeutic effect and mechanism of Oxymatrine on seveve acute pancreatntis (SAP). Methods: Eighty SD rats were randomized into three groups: sham operation(SO) group, NS treatment group, oxymatrine treatment group. SAP model of rats was induced by vetro-injection of 5% sodium tamrocholate into biliopancreatil duct. The rats in the SAP-OM group received OM(30mg/100g)and the SAP-NS group received NS(0.5ml/100g)via the tail vein. 30 minutes before inducing the SAP model. Rats in the three groups were killed at 3,6 hours after induction of the model. The levels of amylase ascites, TNF-α,IL-1β; Resected the pancreatic tissue.The wet/dry ratis of pancreatic were measured. Assessed the integral of the pancreatic tissue, myeloperoxidase (MPO), TNF- α mRNA,IL-1 β mRNA were evaluated. Results: The ascites, the wet/dry ratio of pancreatic, the integral of the pancreatic tissue is belower in SAP-OM group than in SAP-NS group(P〈0.01); The serum levels of TNF- α ,IL-1β is belower in SAP-OM group than in SAP-NS group(P〈0.01);The expression myeloperoxidase (MPO) in pancreatic, TNF-α mRNA, IL-1β mRNA is belower in SAP-OM group than in SAP-NS group(P〈0.01). Conclusions: Oxymatrine benefits the treatment of seveve acute pancreatitis, possibly by decreasing serum TNF-α,IL- 1β level.
