猪血浆免疫球蛋白分离提取技术的研究

摘要 本文用正交试验研究了乙醇沉淀法和硫酸铵盐析法提取免疫球蛋白的条件,简化了传统的工艺流程,比较两种方法的优劣,确定用乙醇沉淀法提取免疫球蛋白。最佳的提取条件是:在乙醇浓度8%,pH7.2,蛋白质浓度5.0%,反应1h后离心去除纤维蛋白原;在乙醇浓度25%,pH6.0,蛋白质浓度2.6%,反应5h沉淀免疫球蛋白。 In this paper, the extraction of immunoglobulin from porcine plasma was studied by orthodoxy experiments and traditional predigested technique. Ethanol and ammonium sulfate were used to extract immunoglobulin. The results suggested that immunoglobulin extraction by ethanol precipitation should he more practical and feasible for its high content, activity and low cost. The optimal conditions were that fibrinogen was deposited at 8% ethanol, pH 7.2, and 5.0% protein to remove fihrinogen, and immunoglobulin was extracted at 25% ethanol, pH 6.0, and 2.6% protein.
