核糖体蛋白基因在胃肠道肿瘤中的表达

摘要 目的探讨核糖体蛋白(RP)基因在胃肠道肿瘤中的表达。方法对NCB I网站中的基因表达系列分析(SAGE)数据库公布的80个癌和癌旁正常组织RP基因进行对比分析(包括结肠癌和胃癌)后,取胃癌、结肠癌标本各30例,通过逆转录聚合酶链反应技术(RT-PCR)对RP L13(RPL13)、RPL37、RPS2和RPS19基因的表达进行检测。结果SAGE数据库分析表明:RPL13、RPL37、RPS19、RPS2和RPLP2在胃癌中表达比正常胃组织中表达平均分别高6.25、2.40、3.86、2.22和3.15倍,在结肠癌中的表达比正常结肠组织中表达平均分别高3.22、7.10、2.55、5.87和2.84倍;RT-PCR结果显示胃癌和结肠癌组织中RPL13、RPL37、RPS2和RPS19 mRNA的表达均高于正常组织(P<0.05)。结论胃肠道肿瘤中存在着共同高表达RP基因,这些共同高表达RP基因可能与消化道肿瘤的发生、发展密切相关。 Objective To explore the gene expression of ribosomal protein(RP) in gastroenteric tumor.Methods 80 samples of RP genes of carcinoma,including 30 samples of colon carcinoma and 30 samples of gastric carcinoma,and adjacent normal tissues in the serial analysis of gene expression(SAGE) library downloaded from NCBI website had a comparative analysis.From the analyzed data,RPL13,RPL37,RPS2 and RPS19 as candidate genes were selected.Those genes of 30 clinical samples of gastric carcinoma and colon carcinoma were determined respectively by reverse transcription polymerase chain reaction(RT-PCR).Results SAGE library data showed that the expressions of RPL13,RPL37,RPS19,RPS2 and RPLP2 genes in gastric carcinoma were 6.25,2.40,3.86,2.22 and 3.15 times more than those in adjacent normal tissues,respectively;the expressions of these RP genes in colon carcinoma were 3.22,7.10,2.55,5.87 and 2.84 times more than those in adjacent normal tissues.RT-PCR results showed that the expressions of RPL13,RPL37,RPS2 and RPS19 from 30 cases of gastric carcinoma and colon carcinoma were significantly higher than those from normal tissues respectively(P〈0.05).Conclusions Some high expressions of RP genes in gastroenteric tumor may have relationship with the genesis and development of gastroenteric tumor.
