摘要 目的研究内皮细胞单核细胞趋化蛋白-1(monocyte chemotacticprotein 1,MCP-1)在颅内动脉瘤壁的表达及其在炎症反应过程中的作用。方法天坛医院2006年10月,至2007年9月术中随机取得9例颅内动脉瘤,以同期5例脑肿瘤患者的颞浅动脉为对照,通过苏木素-伊红(hematoxylin and eosin,HE)染色及Western-blot方法研究MCP-1在颅内动脉瘤发生中所起的作用。结果 HE染色显示,相对于正常动脉壁,动脉瘤其内层、外层网架结构均有破坏;Western-blot显示,MCP-1在动脉瘤中的蛋白表达明显增高。结论 MCP-1在囊性动脉瘤壁中的高表达与动脉瘤的发展有相关性。通过某些途径阻断炎性反应的过程可能防止囊性动脉瘤的形成。 Objective To determine the expression of monocyte chemotactic protein I(MCP-1) in cerebral aneurysms and the role of MCP-1 during this inflammation process. Methods Aneurysm tissues were collected from 2006.10 to 2007.9 at the time of microsurgery from nine patients and compared with control tissues from the superficial temporal arteries of five patients with intracranial tumors. Hematoxylin and eosin(H&E) staining was used to localize and Western-blot analysis was used to determine MCP-1 expression in the different tissues. Results H&E staining showed the inner and outer layered structure of the aneurysm were both destroyed compared with the control artery. MCP-1 was present in the wall of all the aneurysm tissue but was rarely identified in control artery tissue. MCP-1 was identified by immunohistochemistry and western blot using specific antibodies. Conclusion Increased MCP-1 may have deleterious effects on cerebral arteries by promoting inflammation, thus weakening vessel walls and inducing cerebral aneurysms.
