摘要
目的:建立反相高效液相色谱法检测小麦中玉米赤霉烯酮的分析方法。方法:用氯仿提取小麦样品中的玉米赤霉烯酮,以甲醇-水作为流动相,以C18柱分离,并通过荧光检测器检测。结果:该方法的标准曲线回归方程为y=2.97×103+5.23×104x(r=0.9998);小麦样品3个水平的加标回收率分别为85.1%,87.0%和89.5%,相对标准偏差分别为1.02%,3.67%和4.24%;加标浓度为50μg/kg的小麦样品的日内相对标准偏差为2.48%,日间相对标准偏差为4.23%;方法最低检出限为3.0μg/kg。盲样测定结果在给定浓度范围内。结论:该方法灵敏度高、稳定性好、成本低,可以用于小麦中玉米赤霉烯酮的检测。
Objective:To develop a reversed-phase high performance liquid chromatography(HPLC) method for the quantitative determination of zearalenone in wheat.Methods:Zearalenone in wheat samples was extracted with chloroform and quantified by reversed-phase HPLC with fluorometric detector,using methanol-water as mobile phase.Results:The linear equation was y=2.97×103+5.23×104x(r=0.9998).The recoveries of wheat samples spiked with zearalenone were 85.1%,87.0% and 89.5%,and the relative standard deviation(RSD) were 1.02%,3.67% and 4.24%,respectively.The intra-day RSD of spiked wheat was 2.48%,and the inter-day RDS was 4.23%.The detection limit was 3.0 μg/kg.The detections of a blind sample gave results within the given concentration range.Conclusion:This method is highly sensitive and stable with low cost,and can be used for the detection of zearalenone in wheat quantitatively.
